Flow cytometric analysis of CAD expression in HeLa cells. Cells from the Human HeLa (Cervical adenocarcinoma, ATCC® CCL-2™) cell line were fixed with BD Cytofix™ Fixation Buffer (Cat. No. 554655) and permeabilized with BD Phosflow™ Perm Buffer III (Cat. No. 558050). The cells were then stained with either Alexa Fluor™ 647 Rabbit IgG Isotype Control (Cat. No. 569345; dashed line histogram) or Alexa Fluor™ 647 Rabbit Anti-Human CAD antibody (Cat. No. 570746; solid line histogram) at 0.125 µg/test. The fluorescence histogram showing CAD expression (or Ig Isotype control staining) was derived from gated events with the forward and side light-scatter characteristics of intact cells. BD® CompBeads can be used as surrogates to assess fluorescence spillover (compensation). When fluorochrome conjugated antibodies are bound to BD® CompBeads, they have spectral properties very similar to cells. However, for some fluorochromes there can be small differences in spectral emissions compared to cells, resulting in spillover values that differ when compared to biological controls. It is strongly recommended that when using a reagent for the first time, users compare the spillover on cell and BD® CompBeads to ensure that BD® CompBeads are appropriate for your specific cellular application.

- Alexa Fluor® 647 fluorochrome emission is collected at the same instrument settings as for allophycocyanin (APC).
- Since applications vary, each investigator should titrate the reagent to obtain optimal results.
- An isotype control should be used at the same concentration as the antibody of interest.
- Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

The EP710Y recombinant monoclonal antibody specifically recognizes Carbamoyl-phosphate synthetase 2, aspartate transcarbamylase, and dihydroorotase (CAD), which is also known as CAD protein. CAD is a ~243 kDa multifunctional polypeptide with four distinct enzymatic domains: glutamine amidotransferase (GATase), carbamylphosphatesynthetase II (CPSIIase), aspartate transcarbamylase (ATCase), and dihydroorotase (DHOase), which self-assemble into hexamers. In mammalian cells, CAD initiates and controls the first three speed-limiting steps of enzymes carbamoyl-phosphate synthetase, aspartate transcarbamylase and dihydroorotase in the 6-step pathway of pyrimidine synthesis, essential for cell proliferation. The fusion of these first enzymatic activities into a single multifunctional CAD unit is unique to animals. This ubiquitously expressed cytoplasmatic protein is regulated by the mitogen-activated protein kinase (MAPK) cascade. CAD is also involved in protein glycosylation and biosynthesis of phospholipids. Deregulation of CAD activity is associated with tumorigenesis, neurological disorders, and inherited metabolic diseases.
