CD57, clone HNK-1, is derived from hybridization of mouse P3-X63-Ag8.653 myeloma cells with lymph node cells from BALB/c mice immunized with membrane extracts of the HSB-2 T–lymphoblastoid cell line.
CD57 (Leu-7) recognizes an Mr 110-kdalton (kDa) glycoprotein that is expressed on a subset of natural killer (NK) lymphocytes and T lymphocytes. The antigen is a carbohydrate structure associated with myelin-associated glycoprotein (MAG).
FITC, fluorescein isothiocyanate, is a fluorochrome with a molecular weight of 389 Da. FITC is sensitive to pH changes and photobleaching. Due to nearly identical excitation and emission properties but different spillover characteristics, FITC and Alexa Fluor® 488 cannot be used simultaneously. FITC is relatively dim and should be reserved for highly expressed markers whenever possible.

Preparation and Storage
The monoclonal antibody is supplied as 25 µg puriﬁed immunoglobulin in 2.0 mL (12.5 µg/mL) of phosphate-buffered saline. The FITC conjugate is supplied as 25 µg in 2.0 mL (12.5 µg/mL). The biotin conjugate is supplied as 25 µg in 2.0 mL (12.5 µg/mL). Buffered saline contains gelatin and 0.1% sodium azide. The vials should be stored at 2° to 8°C. Conjugated forms should not be frozen and should be protected from prolonged exposure to light. Each reagent is stable for the period shown on the bottle label when stored as directed.
