Purified Mouse Anti-RORγt
Western blot analysis of RORγt expression by mouse thymocytes. Cell lysate was prepared from untreated C57BL/6 mouse thymus (lanes 1-3). The lysate was electrophoresed (SDS-PAGE; approximately 12.5 μg of total cellular protein per lane), transferred to a PVDF membrane and then probed with Purified Mouse Anti-RORγt antibody (Clone O28-835; Cat. No. 562197) at concentrations of 1.25 (lane 1), 0.6 (lane 2), and 0.3 µg/ml (lane 3). Proteins were detected using HRP Goat Anti-Mouse Ig (Cat. No. 554002) and a chemiluminescent detection system. RORgt is identified as a band of ~56 kDa in the thymus cell lysate.

O28-835

The O28-835 monoclonal antibody recognizes ROR-gamma (RORγ) and its isoform, RORγt. RORγ (~58 kDa calculated molecular weight) is a DNA-binding transcription factor that belongs to the ROR/RZR orphan nuclear receptor family. It is expressed in different tissues including skeletal muscle, kidney, liver and adipose tissues. RORγt (~56 kDa calculated molecular weight) is a RORγ isoform that is expressed exclusively by lymphoid cells. It is expressed by various lymphoid cell types including CD4+CD8+ thymocytes, peripheral CD4+ Th17 and CD8+ Tc17 cells and lymphoid tissue inducer (Lti) cells. RORγt plays essential roles in thymopoiesis, T cell homeostasis, differentiation of effector T lymphocytes and the development of secondary lymphoid tissues including lymph nodes and Peyer's patches.

Tissue culture supernatant is purified by either protein A/G or affinity purification methods. Both methods yield antibody in solution that is free of most other soluble proteins, lipids, etc. This format provides pure antibody that is suitable for a number of downstream applications including: secondary labeling for flow cytometry or microscopy, ELISA, Western blot, etc.
