Flow cytometric analysis of Galectin-9 expression by mouse thymocytes. C57BL/6 mouse thymocytes were fixed and permeabilized with BD Cytofix/Cytoperm™ Fixation and Permeabilization Solution (Cat. No. 554722), and stained in BD Perm/Wash™ Buffer (Cat. No. 554723) with either PE Rat IgG2a, κ Isotype Control (Cat. No. 553930; dashed line histogram) or PE Rat Anti-Mouse Galectin-9 antibody (Cat. No. 566027; solid line histogram). The fluorescence histogram showing Galectin-9 expression (or Ig Isotype control staining) was derived from gated events with the forward and side light-scatter characteristics of intact thymocytes. Flow cytometric analysis was performed using a BD FACSCanto™ II Flow Cytometer System.

The RG9-35 monoclonal antibody specifically binds to Galectin 9 (Gal-9) which is encoded by Lgals9 (lectin, galactose binding, soluble 9). Galectin-9 is expressed within cells and can be conveyed to the cell surface and released in soluble form. It is expressed by various cell types including lymphocytes, thymocytes, macrophages, dendritic cells, astrocytes, mast cells, eosinophils, fibroblasts, epithelial cells, and endothelial cells. Its expression can be induced by cells in response to proinflammatory cytokines or infection by certain viruses. Galectin-9 is a ~ 40 kDa S-type lectin that has two carbohydrate recognition domains (CRDs) and forms homodimers. It binds to β-galactosides and can serve as a ligand for TIM-3 (CD366). Galectin-9 can play multiple roles in innate and adaptive immune responses including the activation of macrophage cytokine secretion and bactericidal functions, promotion of dendritic cell maturation, regulatory T cell expansion, and negative regulation of Th1, Th17, NK, and cytotoxic T cells. The RG9-35 antibody can reportedly block some biological activities of Galectin-9.
