BD Pharmingen™ Dog Activated T Lymphocyte Cocktail
(RUO)Three-color analysis of the expression of LSM 8.358 (CD3/Pan T marker), LSM 11.425 (B cell marker) and CTL 2.58 (activation marker) on lysed whole blood from Canine. Dog PBMC were stained with Dog Activated T Lymphocyte Cocktail (Cat. No. 558704). During data analysis, lymphocytes were identified by scatter profile. The figure on the left represents the LSM 8.358 (Pan T marker) and CTL 2.58 (activation marker) profile on resting lymphocytes while the figure in the middle represents these markers on Concanavalin A-stimulated PBMC. The panel on the right represents the profile for LSM 8.58 (Pan T marker) and LSM 11.425 (B cell marker) on Concanavalin A-stimulated cells. Flow cytometry was performed on a BD FACSCalibur™.

The Dog Lymphocyte Activation Cocktail is a three-color reagent cocktail designed to identify dog activated lymphocytes by direct immunofluorescent staining with flow cytometric analysis. The LSM 8.358 antibody reacts with a CD3-like T-cell receptor-associated cell-surface antigen found on thymocytes and peripheral T lymphocytes. The LSM 11.425 antibody was generated against cells derived from canine peripheral lymph nodes and has been used as a prognostic tool in dog B cell lymphoma studies. The CTL 2.58 antibody was generated by using whole cell immunizations of IL-2 dependent feline T cell lines stimulated with PHA and Con A. This clone reacts with dog and is referred to as the T Cell Activation Marker.

BD® CompBeads can be used as surrogates to assess fluorescence spillover (compensation). When fluorochrome conjugated antibodies are bound to BD® CompBeads, they have spectral properties very similar to cells. However, for some fluorochromes there can be small differences in spectral emissions compared to cells, resulting in spillover values that differ when compared to biological controls. It is strongly recommended that when using a reagent for the first time, users compare the spillover on cell and BD® CompBeads to ensure that BD® CompBeads are appropriate for your specific cellular application.

- This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).
- An isotype control should be used at the same concentration as the antibody of interest.
- Source of all serum proteins is from USDA inspected abattoirs located in the United States.
- Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.
