Flow cytometric analysis of CPT1A expression in MCF-7 cells. Cells from the Human MCF-7 (Breast adenocarcinoma, ATCC® HTB-22™) cell line were fixed with BD Cytofix™ Fixation Buffer (Cat. No. 554655) and permeabilized with BD Phosflow™ Perm Buffer III (Cat. No. 558050). The cells were then stained with BD Horizon™ V450 Mouse IgG2b, κ Isotype Control (Cat. No. 560374; dashed line histogram) or BD Horizon™ V450 Rabbit Anti-Human CPT1A antibody (Cat. No. 569344; solid line histogram) at 0.25 µg/test. The fluorescence histogram showing the expression of CPT1A (or Ig Isotype control staining) was derived from gated events with the forward and side-light scatter characteristics of intact cells. Flow cytometry and data analysis were performed using a BD LSRFortessa™ X-20 Cell Analyzer System and FlowJo™ software. Data shown on this Technical Data Sheet are not lot specific.

BD® CompBeads can be used as surrogates to assess fluorescence spillover (compensation). When fluorochrome conjugated antibodies are bound to BD® CompBeads, they have spectral properties very similar to cells. However, for some fluorochromes there can be small differences in spectral emissions compared to cells, resulting in spillover values that differ when compared to biological controls. It is strongly recommended that when using a reagent for the first time, users compare the spillover on cells and BD® CompBeads to ensure that BD® CompBeads are appropriate for your specific cellular application.

- Since applications vary, each investigator should titrate the reagent to obtain optimal results.
- An isotype control should be used at the same concentration as the antibody of interest.
- Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

- BD Horizon V450 has a maximum absorption of 406 nm and maximum emission of 450 nm. Before staining with this reagent, please confirm that your flow cytometer is capable of exciting the fluorochrome and discriminating the resulting fluorescence.

The 8F6AE9 monoclonal antibody specifically recognizes Carnitine palmitoyltransferase 1A (CPT1A), also known as, Carnitine O-palmitoyltransferase 1, liver isoform. CPT1A is a ~ 88 kDa enzyme encoded by CPT1A which belongs to the carnitine/choline acetyltransferase family. CPT1A is located in the mitochondrial outer membrane but facilitates carnitine-dependent transport of fatty acids across the inner mitochondrial membrane and plays a key role in triglyceride metabolism. CPT1A is the primary isoform and is found in the liver, spleen, kidneys, lungs, intestines, pancreas, brain and ovaries. Overexpression of CPT1A has been reported in tumor cell lines and its deficiency is associated with many metabolic disorders. Recently, CPTIA has been suggested as a novel prognostic indicator and potential target for anti-cancer therapy.
