Preparation and Storage

The monoclonal antibody is supplied as 10 μg purified immunoglobulin in 2.0 mL (5 μg/mL) of PBS. The FITC conjugate is supplied as 20 μg in 2.0 mL (10 μg/mL) of PBS. The PE conjugate is supplied as 3 μg in 1.0 mL (3 μg/mL) of PBS. The PE-Cy7 conjugate is supplied as 6.25 μg in 0.5 mL (12.5 μg/mL) of PBS. The APC conjugate is supplied as 6 μg in 0.5 mL (12 μg/mL) of PBS. PBS contains gelatin and 0.1% sodium azide.

The CD25 antibody, clone 2A3. is derived from the hybridization of NS-1 mouse myeloma cells with spleen cells isolated from BALB/c mice immunized with PHA-activated human T lymphocytes.

The CD25 antibody recognizes a 55-kilodalton (kDa) type I integral membrane glycoprotein. The CD25 antigen is also known as the human low-affinity interleukin-2 receptor α chain (IL-2Rα) and the Tac antigen.

PE-Cy7 dye is a part of the BD PE family of dyes. This tandem fluorochrome is comprised of a R-Phycoerythrin (PE) donor that has excitation maxima (Ex Max) of 496-nm and 566-nm and an acceptor dye, Cy™7, with an emission maximum (Em Max) at 781-nm. PE can be excited by the Blue (488-nm), Green (532-nm) and yellow-green (561-nm) lasers and detected using an optical filter centered near 781 nm (e.g., a 760/60-nm bandpass filter). The donor dye can be excited by the Blue (488-nm), Green (532-nm) and yellow-green (561-nm) lasers and the acceptor dye can be excited by the Red (627–640-nm) laser resulting in cross-laser excitation and fluorescence spillover. Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
