Two-parameter flow cytometric analysis of TLR6 (CD286) expression on human peripheral blood leucocyte populations. Human peripheral blood cells were first treated with BD Pharm Lyse™ Lysing Buffer (Cat. No. 555899) to lyse erythrocytes. The leucocytes were then washed and stained with either PE Mouse IgG1, κ Isotype Control (Cat. No. 559320; Left Plot) or PE Mouse Anti-Human TLR6 (CD286) antibody (Cat. No. 566339; Right Plot). Two-parameter flow-cytometric contour plots showing the correlated expression of TLR6 (CD286) [or Ig Isotype control staining] versus side light-scatter signals were derived from gated events with the forward and side light-scatter characteristics of viable human peripheral blood leucocytes. Flow cytometric analysis was performed using a BD LSRFortessa™ X-20 Flow Cytometer System.

- This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).
- An isotype control should be used at the same concentration as the antibody of interest.
- Source of all serum proteins is from USDA inspected abattoirs located in the United States.
- Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

The TLR6.127 monoclonal antibody specifically recognizes Toll-like receptor 6 (TLR6), which is also known as CD286. CD286 belongs to the mammalian Toll-like receptor (TLR) family of pattern recognition receptors that participate in inducing innate and adaptive immune responses to microbial pathogens. TLR6 is a type I transmembrane glycoprotein comprised of an extracellular domain with a large number of leucinerich repeats (LRRs). Its cytoplasmic region contains a signaling Toll/Interleukin-1 Receptor (TIR) domain. TLR6 is expressed on monocytes, neutrophils, and some dendritic cells. In association with TLR2 (CD282), TLR6 recognizes diacylated lipopeptides and peptidoglycans of bacterial or fungal origin. The ligand-bound TLR2:TLR6 complex transduces responses intracellularly through MyD88 and the NF-κB signaling cascade leading to the production of proinflammatory cytokines by responding cells.
