Flow cytometric analysis of mouse GITR Ligand expression on non-transfected and GITR Ligand-transfected cells. Non-transfected (Left Panel) and GITR Ligand-transfected (Right Panel) P815 cells were stained with either Alexa Fluor® 647 Rat IgG2b, κ Isotype Control (Cat. No. 557691) or Alexa Fluor® 647 Rat Anti-Mouse GITR Ligand antibody (Cat. No. 563542). Fluorescence histograms showing the expression of GITR Ligand (solid line histograms) or Ig Isotype control staining (dashed line histograms) on non-transfected or GITR Ligand-transfected P815 cells were derived from gated events with the forward and side light-scatter characteristics of viable cells. Flow cytometric analyses were performed using a BD™ LSR II Flow Cytometer System.

- An isotype control should be used at the same concentration as the antibody of interest.
- Since applications vary, each investigator should titrate the reagent to obtain optimal results.

The MIH44 monoclonal antibody specifically binds to Glucocorticoid-induced TNF-related Ligand (GITR Ligand). The GITR Ligand protein is encoded by the Tnfsf18 (Tumor necrosis factor ligand superfamily member 18) gene. The GITR Ligand is a type II transmembrane protein that is expressed on endothelial cells and antigen presenting cells, including B cells, macrophage and dendritic cells. The GITR Ligand reportedly costimulates T cell activation and reverses CD4+CD25+ Treg cell-mediated suppression by interaction with its cognate receptor, GITR that is expressed by these cells.
