Two-color flow cytometric analysis of IgM expression on mouse splenocytes. BALB/c mouse splenic leucocytes were preincubated with Purified Rat Anti-Mouse CD16/CD32 (Mouse BD Fc Block™) (Cat. No. 553141/553142). The cells were then stained with PE-CF594 Hamster Anti-Mouse CD3e (Cat. No. 562286/562332) and either BUV395 Rat IgG2a, κ Isotype Control (Cat. No. 563556, Left Panel) or BUV395 Rat Anti-Mouse IgM antibody (Cat. No. 564025, Right Panel). Fluorescence two-color dot plots showing the correlated expression of IgM (or Ig Isotype control staining) versus CD3e were derived from gated events with the light-scattering characteristics of viable splenocytes. Flow cytometric analysis was performed on a BD™ LSR II.

For optimal and reproducible results, BD Horizon Brilliant™ Stain Buffer should be used anytime BD Horizon Brilliant™ dyes are used in a multicolor flow cytometry panel. Fluorescent dye interactions may cause staining artifacts which may affect data interpretation. The BD Horizon Brilliant Stain Buffer was designed to minimize these interactions. When BD Horizon Brilliant Stain Buffer is used in in the multicolor panel, it should also be used in the corresponding compensation controls for all dyes to achieve the most accurate compensation. For the most accurate compensation, compensation controls created with either cells or beads should be exposed to BD Horizon Brilliant Stain Buffer for the same length of time as the corresponding multicolor panel. The R6-60.2 antibody monoclonal antibody specifically binds to mouse Immunoglobulin M (IgM) of Igh-C[a] and Igh-C[b] haplotypes. It does not react with other Ig isotypes. The R6-60.2 antibody has not been shown to stimulate B-cell proliferation.
