Description
This kit enables single-cell Whole Transcriptome Analysis (WTA) library preparation from cells captured on the BD Rhapsody™ HT Xpress System, supporting both standalone WTA workflow and the multiomic application that combines WTA with BD® AbSeq for CITE-seq analysis. The kit accommodates sample multiplexing using any of the BD® Single-Cell Multiplexing Kits across both configurations, whether using WTA alone or in combination with BD® AbSeq. For specialized multiomic assays such as ATAC-Seq or TCR/BCR profiling, this kit provides the WTA library preparation components while additional assay-specific reagents must be purchased separately. The kit provides a set of 8 forward index primers and 16 reverse index primers—8 optimized for WTA and 8 for BD® AbSeq/multiplexing applications—enabling unique dual-indexed library preparation. Store all BD Omics One Whole Transcriptome Analysis (WTA) v3.0 kit components at -25 °C to -15 °C.
Recommended Assay Procedures
After retrieval and cDNA synthesis of cell capture beads from the BD Rhapsody™ 8-Lane Cartridge, WTA products are made by first random priming and extension followed by amplification. BD® AbSeq and Sample Tag molecules, if present, are denatured and amplified using the included Sample Tag and BD® AbSeq primers. Indexes for Next Generation Sequencing (NGS) libraries are included for up to 8 WTA and associated multiomic libraries.
Designed for use on 1,000 – 100,000 cells per test.

Danger: PCR Master Mix (51-9025466) contains 5-10 % tris base, CAS number 77-86-1, EC number 201-064-4 and 1-5 % tetramethylammonium chloride, CAS number 75-57-0, EC number 200-880-8. These reagents are classified as hazardous according to the Globally Harmonized System of Classification and Labelling of Chemicals (GHS).
