Two color flow cytometric analysis of Platelet GPVI expression on human peripheral blood platelets. Platelets were isolated from fresh whole blood and fixed with BD Cytofix™ Fixation Buffer (Cat. No. 554655). After washing, the fixed platelets were stained with FITC CD41a (Cat.No. 555466/557296/56185) and either Alexa Fluor® 647 Mouse IgG1, κ Isotype Control (Cat.No.557732; Left Panel) or Alexa Fluor® 647 GPVI antibody (Cat. No. 564701; Right Panel). Two-color flow cytometric contour plots showing the correlated expression of Platelet GPVI (or Ig isotype control staining) versus CD41a were derived from events with the forward and side light-scatter characteristics of platelets. Flow cytometric analysis was performed using a BD LSRFortessa™ Cell Analyzer System.

Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated to Alexa Fluor® 647 under optimum conditions, and unreacted Alexa Fluor® 647 was removed.

  1. This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).

  7. An isotype control should be used at the same concentration as the antibody of interest.
  8. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

HY101

The HY101 monoclonal antibody specifically binds to a collagen receptor, Platelet Glycoprotein VI (GPVI or GP6). GPVI is a 58-kD platelet membrane glycoprotein that belongs to the immunoglobulin superfamily. Upon blood vessel wall injury and consequent endothelial cell damage, platelets collect upon the exposed collagen fibers present in the subendothelial matrix. The platelet and collagen interaction leads to activated platelet aggregation and thrombus formation. Although normally serving a protective function in clot formation that limits blood loss, the platelet GPVI/collagen interaction may also play a role in myocardial infarction and stroke which are associated with the rupture of atherosclerotic plaques.

Alexa Fluor™ 647 Dye is part of the BD red family of dyes. This is a small organic fluorochrome with an excitation maximum (Ex Max) at 653-nm and an emission maximum (Em Max) at 669-nm. Alexa Fluor 647 is designed to be excited by the Red laser (627-640 nm) and detected using an optical filter centered near 520-nm (e.g., a 660/20 nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
