Preparation And Storage

The PE conjugate is supplied as 10 μg in 1.0 mL (10 μg/mL) of phosphate-buffered saline (PBS). The PerCP-Cy5.5 conjugate is supplied as 1 μg in 1.0 mL (1 μg/mL) of PBS. The PE-Cy7 conjugate is supplied as 6.25 μg in 0.5 mL (12.5 μg/mL) of PBS. The APC conjugate is supplied as 5 μg in 0.5 mL (10 μg/mL) of PBS. PBS contains gelatin

and 0.1% sodium azide.

Store vials at 2° to 8°C. Conjugated forms should not be frozen and should be protected from prolonged exposure to light. Each reagent is stable for the period shown on the bottle label when stored as directed.

The CD117 antibody, clone 104D2, is derived from the hybridization of Sp2/0 mouse myeloma cells with spleen cells isolated from BALB/c mice immunized with the megakaryocytic cell line MOLM-1.

The CD117 antibody binds to a 145-kilodalton (kDa) type I transmembrane glycoprotein in the receptor tyrosine kinase (RTK) family. The CD117 antigen is also known as c-kit and stem cell factor receptor (SCFR).

PerCP-Cy5.5 dye is part of the BD blue family of dyes. This tandem fluorochrome is comprised of a fluorescent protein complex (PerCP) with an excitation maximum (Ex Max) of 482 nm and an acceptor dye with an emission maximum (Em Max) at 676 nm. PerCP-Cy5 is designed to be excited by the blue laser (488-nm) and detected using an optical filter centered near 680 nm (e.g., a 695/40 nm bandpass filter). The donor dye can be partially excited by the Violet (405-nm) laser resulting in cross-laser excitation and fluorescence spillover. Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
