APC-R700 Rat Anti-Mouse CD200
Multicolor flow cytometric analysis of CD200 expression on mouse thymocytes. Mouse thymocytes were preincubated with Purified Rat Anti-Mouse CD16/CD32 antibody (Mouse BD Fc Block™) (Cat. No. 553141/553142). The cells were then stained with FITC Rat Anti-Mouse CD4 (Cat. No. 553047/553046/561835) and PE Rat Anti-Mouse CD8a (Cat. No. 553033/553032/561095) antibodies, and either BD Horizon™ APC-R700 Rat IgG2a, κ Isotype Control (Cat. No. 564982; dashed line histograms) or BD Horizon™ APC-R700 Rat Anti-Mouse CD200 antibody (Cat. No. 565546; solid line histograms). The fluorescence histograms showing CD200 expression (or Ig Isotype control staining) were derived from CD4-CD8a+ (Top Left Plot ), CD4+CD8a+ (Top Right Plot), CD4-CD8a- (Bottom Left Plot), or CD4+CD8a- (Bottom Right Plot) gated events with the forward and side light-scatter characteristics of viable thymocytes. Flow cytometric analysis was performed using a BD LSRFortessa™ Cell Analyzer System.

Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with BD Horizon APC-R700 under optimum conditions, and unconjugated antibody and free BD Horizon APC-R700 were removed.

OX-90

The OX-90 monoclonal antibody specifically recognizes CD200 (OX-2 antigen), a type-1 transmembrane glycoprotein containing two extracellular Ig-like domains. CD200 is expressed on thymocytes, neurons, B lymphocytes, splenic follicular dendritic cells and endothelium, and subsets of T lymphocytes and dendritic cells; but not on NK cells, granulocytes, monocytes, or macrophages. CD200 Receptor (CD200R; OX2R) expression is restricted to myeloid cells, and it appears that its engagement with CD200 results in inhibition and/or down-regulation of myeloid cell activity.

This antibody was conjugated to BD Horizon APC-R700, which has been developed exclusively by BD Biosciences as a better alternative to Alexa Fluor® 700. APC-R700 excites and emits at similar wavelengths to Alexa Fluor® 700 yet exhibits significantly improved brightness. This dye can be excited by the red laser and detected with the same filter set as Alexa Fluor® (eg, 730/45-nm filter).

The BD Horizon™ APC-R700 (APC-R700) Dye is a part of the BD APC red family of dyes. This tandem fluorochrome is comprised of an Allophycocyanin (APC) dye donor that has excitation maximum (Ex Max) of 651-nm and an acceptor dye, R700, with an emission maximum (Em Max) at 706-nm. APC-R700, driven by BD innovation, is designed to be excited by the red (627–640-nm) laser and detected using an optical filter centered near 710-nm (e.g., a 720/40-nm bandpass filter). APC-R700 is a brighter alternative to Alexa Fluor™ 700. Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
