Flow cytometric analysis of CD26 expression on human peripheral blood lymphocytes. Whole blood was stained with either APC Mouse IgG1, κ Isotype Control (Cat. No. 554681; dashed line histogram) or APC Mouse Anti-Human CD26 antibody (Cat. No. 563670; solid line histogram). Erythrocytes were lysed with BD FACS Lysing Solution (Cat. No. 349202). The fluorescence histograms were derived from gated events with the forward and side light-scatter characteristics of intact lymphocytes. Flow cytometric analysis was performed using a BD™ LSR II Flow Cytometer System.

Note: For best staining results, cell samples should be stained and maintained at 4-8°C before flow cytometric analysis.
BD® CompBeads can be used as surrogates to assess fluorescence spillover (compensation). When fluorochrome conjugated antibodies are bound to BD® CompBeads, they have spectral properties very similar to cells. However, for some fluorochromes there can be small differences in spectral emissions compared to cells, resulting in spillover values that differ when compared to biological controls. It is strongly recommended that when using a reagent for the first time, users compare the spillover on cell and BD® CompBeads to ensure that BD® CompBeads are appropriate for your specific cellular application.

The M-A261 monoclonal antibody specifically binds to CD26. CD26 is also known as dipeptidyl peptidase IV (DPPIV/DPP IV, DPP4) or adenosine deaminase binding protein (ADABP). CD26 is a 110 kDa type II transmembrane glycoprotein. CD26 is expressed by T lymphocytes and increases upon activation. It is also expressed by B cells, NK cells, and macrophages. CD26 is an ectoenzyme which plays an integral role in T-cell proliferation and may interact with extracellular matrix proteins such as fibronectin or collagen.
Allophycocyanin (APC), is part of the BD family of phycobiliprotein dyes. This fluorochrome is a multimeric fluorescent phycobiliprotein with excitation maximum (Ex Max) of 651 nm and an emission maximum (Em Max) at 660 nm. APC is designed to be excited by the Red (627-640 nm) laser and detected using an optical filter centered near 660 nm (e.g., a 660/20 nm bandpass filter). Please ensure that your instrument's configurations (lasers and optical filters) are appropriate for this dye.
The CD26 antigen can serve as a receptor for the coronavirus, MERS-CoV.
