Multicolor flow cytometric analysis of KIR2DS4 (CD158i) expression on human peripheral blood lymphocytes. Whole blood was stained with PE Mouse Anti-Human CD56 (Cat No. 555516/561903) and FITC Mouse Anti-Human CD3 (Cat No. 555332/561806/561807) antibodies, and either Alexa Fluor® 647 Mouse IgG2a, κ Isotype Control (Cat. No. 557715; Left Plot) or Alexa Fluor® 647 Mouse Anti-Human KIR2DS4 (CD158i) antibody (Cat. No. 564375; Middle and Right Plots). Erythrocytes were lysed with BD Pharm Lyse™ Lysing Buffer (Cat. No. 555899). The two-color flow cytometric contour plots show the correlated expression patterns of CD56 (Left and Middle Plots) or CD3 (Right Plot) versus KIR2DS4 (CD158i) [or Ig Isotype control staining] for gated events with the forward and side light-scatter characteristics of viable peripheral blood lymphocytes. Flow cytometric analysis was performed using a BD™ LSR II Flow Cytometer System.

- This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).

- Alexa Fluor® 647 fluorochrome emission is collected at the same instrument settings as for allophycocyanin (APC).
- An isotype control should be used at the same concentration as the antibody of interest.
- Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.
- Source of all serum proteins is from USDA inspected abattoirs located in the United States.

The 179315 monoclonal antibody specifically binds to Killer cell immunoglobulin-like receptor 2DS4 which is also known as CD158i, KKA3, and NKAT8. CD158i is encoded by KIR2DS4 (killer cell immunoglobulin-like receptor, two domains, short cytoplasmic tail, 4). It is a type I transmembrane glycoprotein that can bind to HLA-C antigens and function as an activating immunoglobulin-like receptor that is expressed on NK cells and some T cells.
