Recommended Assay Procedures
ELISA Capture: The purified C3-4 antibody (Cat. No. 550644) is useful as a capture antibody for a sandwich ELISA for specifically measuring rat IL-6 protein levels. Purified C3-4 antibody can be paired with the biotinylated G307-2 antibody (Cat. No. 550642) as the detection antibody and with recombinant rat IL-6 protein (Cat. No. 557008) as the standard. The purified C3-4 capture antibody should be titrated between 2 - 8 µg/ml to determine optimal concentration for ELISA capture. To obtain linear standard curves, doubling dilutions of rat IL-6 ranging from ~10,000 to 78 pg/ml are recommended for inclusion in each ELISA plate. For specific methodology please visit the protocols sections or the chapter on ELISA in the Immune Function Handbook, both of which are posted on our web site,w ww.bdbiosciences.com.
Note 1: This ELISA pair is recommended primarily for measuring cytokine from experimental cell culture systems. These ELISA reagents are not recommended for assaying serum or plasma samples. For measuring rat IL-6 in serum or plasma our rat IL-6 BD OptEIA™ ELISA Set (Cat. No. 550319) is especially formulated and recommended.
Note 2: This ELISA pair shows no cross-reactivity with rat IL-2, IL-4, IL-10, IFNγ, GM-CSF, TNF, mouse or human IL-6.
Product Notices
- Since applications vary, each investigator should titrate the reagent to obtain optimal results.
- Please refer to www.bdbiosciences.com/us/s/resources for technical protocols.
- Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.
Companion Products
The C3-4 antibody reacts with rat interleukin-6 (IL-6). The immunogen used to generate the C3-4 hybridoma was recombinant rat IL-6.
