Description
3,3' Diaminobenzidine (DAB) is a substrate for the enzyme peroxidase and is a widely used chromogen for immunohistochemical staining and immunoblotting. In the presence of peroxidase, DAB produces a brown precipitate that is insoluble in alcohol.
Preparation And Storage
Recommended Assay Procedures
Immunohistochemistry: Just before staining, add one drop (~ 40 µL) of DAB Chromogen for every 1 mL of DAB Buffer. The mixed DAB substrate solution can be stored for up to 6 hours, making it ideal for use in automated stainers. Storing the reagent for longer periods of time will cause precipitate formation. Apply the mixed DAB substrate reagent to the tissue section after it has been incubated with the appropriate horseradish peroxidase (HRP) conjugated reagents and washing is complete. Approximately 100 µL of the mixed DAB substrate reagent should be sufficient to cover the entire tissue section. Incubate the section with the mixed DAB reagent for 5-15 minutes or until the desired color intensity is obtained.
DAB Buffer (component 51-7548KC) is a 50 mL solution of citric acid and disodium hydrogenorthophosphate.
Danger: DAB Chromogen (component 51-7549KC) is a 2.5 mL solution containing 1.74% 3,3' diaminobenzidine (w/w).
