Multicolor flow cytometric analysis of mouse CD371 (Clec12A) expression on spleen dendritic cell subsets. BALB/c splenocytes were simultaneously stained with V450 Rat Anti-Mouse CD45R (Cat. No. 560472), PerCP-Cy™5.5 Hamster Anti-Mouse CD11c (Cat. No. 560584), Alexa Fluor® 488 Hamster Anti-Mouse CD3e (Cat. No. 557666), APC-H7 Rat Anti-Mouse CD19 (Cat. No. 560245), APC Rat Anti-Mouse CD49b (Cat. No. 560628) and either PE Rat IgG2a, κ Isotype Control (Cat. No. 553930, dashed line histogram) or PE Rat Anti-Mouse CD371 (Clec12A) (Cat. No. 562773, solid line histogram). Overlapping histograms show the levels of CD371 or Ig Isotype control staining in CD11c-high CD45R-negative (Left Panel) and CD11c-intermediate CD45R-positive (Right Panel) populations. Fluorescence histograms were derived from gated CD3- CD19- CD49b- events with the forward and side light-scattering characteristics of viable splenocytes cells. Flow cytometry was performed on a BD™ LSR II.

Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with R-PE under optimum conditions, and unconjugated antibody and free PE were removed.

Product Notices

  1. Since applications vary, each investigator should titrate the reagent to obtain optimal results.
  2. An isotype control should be used at the same concentration as the antibody of interest.
  3. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

5D3/CD371

The 5D3/CD371 (aka, 1/06-5D3) monoclonal antibody specifically binds to mouse CD371 which is encoded by Clec12A (C-type lectin domain family 12 member A). CD371 is also known as Myeloid inhibitory C-type lectin-like receptor (MICL), C-type lectin-like molecule 1 (CLL-1) or Dendritic cell-associated lectin 2 (DCAL-2). CD371 is a ~30 kDa type II transmembrane glycoprotein that possesses one extracellular C-type lectin domain and one cytoplasmic immunoreceptor tyrosine-based inhibitory motif (ITIM). CD371 is expressed on plasmacytoid dendritic cells, subsets of myeloid dendritic cells, monocytes, macrophages and B cells. It reportedly serves as a negative regulator of granulocyte and monocyte function.

R-Phycoerythrin (PE), is part of the BD family of Phycobiliprotein dyes. This fluorochrome is a multimeric fluorescent phycobiliprotein with excitation maximum (Ex Max) of 496 nm and 566 nm and an emission maximum (Em Max) at 576 nm. PE is designed to be excited by the Blue (488 nm), Green (532 nm) and Yellow-Green (561 nm) lasers and detected using an optical filter centered near 575 nm (e.g., a 575/26-nm bandpass filter). As PE is excited by multiple lasers, this can result in cross-laser excitation and fluorescence spillover on instruments with various combinations of Blue, Green, and Yellow-Green lasers. Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
