Performance
Gene expression patterns are preserved in PBMC samples stored in BD OMICS-Guard™ CRYO PB at −80 °C over time. Gene expression levels (A) and sample and cell clustering patterns (B) were assessed in freshly processed samples and compared to samples stored for 10 and 60 days in BD OMICS-Guard™ CRYO PB. Gene detection comparisons across time display highly correlated expression levels with limited impact shown in sample and cell-type clustering data.
Cell-type proportions are preserved in PBMC samples stored in BD OMICS-Guard™ CRYO PB at −80 °C over time. Cell-type composition was assessed in freshly processed samples and compared to samples stored for 10 and 60 days. Following sequencing and data analysis, stored samples showed less than 10% variation in cell-type proportions relative to fresh controls, demonstrating compositional stability across both timepoints.
Gene and protein expression sensitivity are preserved in PBMC samples stored in BD OMICS-Guard™ CRYO PB at −80 °C. Gene expression (A) and protein expression (B) performance metrics were assessed in freshly processed samples and compared to samples stored in BD OMICS-Guard™ CRYO PB for 60 days. Following sequencing and data analysis, stored samples demonstrated equivalent sensitivity across both measures, indicating performance is maintained even after long-term storage.
Representative sample biology and assay performance are conserved in whole blood samples stored in cryopreservation conditions (in BD OMICS-Guard™ CRYO PB) for 3 months. Whole blood samples were either stored directly in BD OMICS-Guard™ CRYO PB or prepared into PBMCs and processed fresh. Following sequencing and data analysis, when stored sample’s metrics were compared to fresh processed samples, gene expression displayed a Pearson correlation value of 0.94 (A) and cell-type specific protein (B) and UMAP clustering patterns remained consistent.
Data from samples processed after storage in BD OMICS-Guard™ CRYO PB were compared to aliquots of the same samples processed fresh in both the BD Rhapsody™ TCR/BCR Next and Multiomic ATAC-Seq Assays. Performance data for TCR/BCR pairing efficiency (A) and UMAP clustering overlays (B) illustrate no reduction in metrics or significant impact compared to fresh samples.
