Description
CD3, clone UCHT1, is derived from hybridization of mouse NS-1 myeloma cells with spleen cells from BALB/c mice immunized with human thymocytes. Anti-MPO, clone 5B8, is generated from the fusion of X63.Ag8-653 myeloma cells with spleen cells from BALB/c mice immunized with commercially purified myeloperoxidase. CD79a, clone HM47, is generated from the fusion of Sp2/0 myeloma cells with spleen cells from BALB/c mice immunized with a synthetic peptide conjugated to thyroglobulin. The peptide had the sequence representing amino acids 202 to 216 inclusive as deduced from the human mb-1 cDNA sequence. CD3 reacts with the epsilon chain of the CD3 antigen/T-cell antigen receptor (TCR) complex. The antigen recognized by CD3 antibodies is noncovalently associated with either α/β or γ/δ TCR, 70 to 90 kilodaltons (kd). Anti-MPO reacts with intracellular MPO, a hemoprotein consisting of two molecular subunits, 60 kd and 12 kd. MPO is a major enzyme involved in the inflammatory responses of polymorphonuclear leucocytes. It catalyzes the production of hypochlorous acid, a potent microbicidal agent. CD79a recognizes the cytoplasmic domain of a 47-kd membrane glycoprotein present on B lymphocytes.
Preparation And Storage
Store vials at 2° to 8°C. Conjugated forms should not be frozen and should be protected from prolonged exposure to light. Each reagent is stable for the period shown on the bottle label when stored as directed.
