Alexa Fluor® 488 Rat Anti-Mouse CD125
Flow cytometric analysis of Alexa 488 rat anti-mouse CD125 on mouse bone marrow cells.  Isolated murine bone marrow cells were stained with either Alexa 488 rat anti-mouse CD125 (clone T21, Cat. No. 558533, right panel) or a Alexa 488 rat IgG1 isotype control (Cat. No. 557720, left panel) and PE rat anti-mouse CD11b (clone M1/70, Cat. No. 553311).  Flow cytometry was performed on a BD FACSCalibur™ instrument and the histograms were derived from the gated events based on light scattering characteristics of viable bone marrow cells.

Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated to Alexa Fluor® 488 under optimum conditions, and unreacted Alexa Fluor® 488 was removed.

Recommended Assay Procedures

Flow cytometry:  For staining of cell suspensions from peripheral lymphoid tissues, it is recommended that the cells be pre-incubated with Purified Rat Anti-Mouse CD16/CD32 (Mouse BD Fc Block™) mAb, Clone 2.4G2 (MN 553141).

  1. Since applications vary, each investigator should titrate the reagent to obtain optimal results.
  2. Alexa Fluor® 488 fluorochrome emission is collected at the same instrument settings as for fluorescein isothiocyanate (FITC).
  3.   4. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

T21

The T21 monoclonal antibody specifically binds to CD125 which is also known as the alpha subunit of the IL-5 Receptor (IL5RA/IL-5Rα).  CD125 is a type I transmembrane protein that belongs to the hemopoietin receptor and Ig gene superfamilies. It is expressed on B cells and eosinophils. CD125 associates with the Common β chain (βc/Bc) subunit, also known as CD131, to form the functional IL-5 Receptor complex. Mouse IL-5 belongs to the hematopoietic growth factor family of cytokines and promotes the growth and differentiation of B cells and eosinophils. Upon binding, the T21 antibody blocks IL-5 binding to its receptor and can inhibit IL-5 induced B-cell proliferation and antibody production.

Alexa Fluor™ 488 Dye is part of the BD blue family of dyes. This is a small organic fluorochrome with an excitation maximum (Ex Max) at 494-nm and an emission maximum (Em Max) at 517-nm. Alexa Fluor™ 488 is designed to be excited by the Blue laser (488 nm) and detected using an optical filter centered near 520-nm (e.g., a 530/30-nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
