To support advanced assay development, BD special order instrumentation deliver the latest in laser technology to researchers to achieve a wide range of color, wavelength and power choices. 

 

The special-order systems are designed and configured in collaboration with customers to meet exact research requirements.

Measurement of the antigen expression of human bone marrow cells

Seven-laser excitation of seven fluorochromes is shown (right), demonstrating the utility of a seven-laser special order BD® LSR II Analyzer.*

 

Each spatially separated beam provides discrete fluorochrome excitation so that virtually no computer compensation is required for complete cell population separation. *The BD® LSR product family includes the special order BD® LSR II and BD LSRFortessa™ Analyzers. This experiment was run on a BD® LSR II Analyzer configured with the following lasers: 20-mW 355-nm UV, 50-mW 405-nm violet, 20–100 mW 488-nm blue (variable power), 150-mW 532-nm green (variable power), 50-mW 594-nm yellow, 40-mW 638-nm red, and 25-mW 785-nm IR.

Probability state model of human B-cell maturation

Human bone marrow was stained with CD23 Pacific Blue™, CD20 AmCyan, TdT FITC, CD34 PerCP, CD43 PE, CD19 PE-Cy5.5, CD10 PE-Cy7, Kappa APC, CD45 APC-Cy7, and Lambda Alexa Fluor™ 594. Approximately 10,000 cells were analyzed. The percentage of cells in each state was plotted against the level of antigen expression measured. B-cell precursors are seen to be downregulating early markers such as CD34 and TdT and upregulating more mature antigens such as CD20, CD23 and surface immunoglobulin light chain.
