Preparation And Storage

The monoclonal antibody is supplied as 25 µg purified immunoglobulin in 2.0 mL (12.5 µg/mL) of phosphate-buffered saline. The FITC conjugate is supplied as 25 µg in 2.0 mL (12.5 µg/mL). The biotin conjugate is supplied as 25 µg in 2.0 mL (12.5 µg/mL). Buffered saline contains gelatin and 0.1% sodium azide. The vials should be stored at 2° to 8°C. Conjugated forms should not be frozen and should be protected from prolonged exposure to light. Each reagent is stable for the period shown on the bottle label when stored as directed.

HNK-1

The CD57 antibody, clone HNK-1, is derived from the hybridization of P3-X63-Ag8.653 mouse myeloma cells with lymph node cells isolated from BALB/c mice immunized with membrane extracts of the HSB-2 T–lymphoblastoid cell line.

The CD57 (Leu-7) antibody recognizes a 110-kilodalton (kDa) glycoprotein that is expressed on a subset of natural killer (NK) lymphocytes and T lymphocytes. The antigen is a carbohydrate structure associated with myelin-associated glycoprotein.

Biotin
Biotin is a ubiquitous co-factor (also known as Vitamin B7) that has many properties that make it extremely useful for molecular biology. Biotin has an extremely high affinity for the Avidin family of proteins (Kd = 10-15 M), making it the perfect tool to link two molecules. Biotin labeled antibodies can be combined with any number of Avidin-conjugated probes in order to customize an assay to a particular need. This is especially useful in the case of magnetic cell separation using streptavidin/magnetic bead conjugates, or in the case of flow cytometry using streptavidin/fluorophore conjugates.
