BD® OMICS-One Dual Index Kit

The dual index kit is designed for multiplexing single-cell libraries in preparation for sequencing. The kit contains the following components:

•  Forward primers: 8 unique primers, FP each with a distinct 8-base index sequence

   –  FP1 Sequence: TATAGCCT

   –  FP2 Sequence: ATAGAGGC

   –  FP3 Sequence: CCTATCCT

   –  FP4 Sequence: GGCTCTGA

   –  FP5 Sequence: AGGCGAAG

   –  FP6 Sequence: TAATCTTA

   –  FP7 Sequence: CAGGACGT

   –  FP8 Sequence: GTACTGAC

•  Reverse primers: 8 unique primers, each with a distinct 8-base index sequence

   –  RP1 Sequence: ATTACTCG

   –  RP2 Sequence: TCCGGAGA

   –  RP3 Sequence: CGCTCATT

   –  RP4 Sequence: GAGATTCC

   –  RP5 Sequence: ATTCAGAA

   –  RP6 Sequence: GAATTCGT

The purpose of the kit is to enable dual indexing of single-cell libraries. Dual indexing involves appending a unique combination of forward and reverse index sequences to each library. This allows for multiple libraries to be pooled and sequenced together, increasing the efficiency of sequencing runs.

The kit provides eight unique dual-index combinations or 64 combinatorial dual-index combinations (8 forward × 8 reverse), supporting the multiplexing of up to 64 single-cell libraries. The primers are designed for compatibility with the BD Rhapsody™ single-cell library preparation protocols.
