Preparation And Storage

The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with biotin under optimum conditions, and unreacted biotin was removed. Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze.

Recommended Assay Procedures

ELISA Capture (Human/Viral IL-10): Biotinylated- JES3-12G8 (Cat. No. 554599) is useful as an ELISA capture antibody when used in combination with purified JES3-9D7 (Cat No. 554497) and with recombinant human IL-10 protein standard (Cat. No. 554611).

Note 1: This ELISA pair shows no cross-reactivity with any of the cytokines tested (e.g., mouse IL-1β, IL-2, IL-3, IL-4, IL-5, IL-6, IL-7, IL-9, IL-10, IL-12 p70, IL-15, GM-CSF, IFN-γ, MCP-1, TCA-3, TNF; human IL-1α, IL-1β, IL-2, IL-3, IL-4, IL-5, IL-6, IL-7, IL-8, IL-9, IL-11, IL-12 p70, IL-12 p40, IL-13, IL-15, G-CSF, GM-CSF, IFN-γ, lymphotactin, MCP-1, MCP-2, MIP-1α, MIP-1β, NT-3, PDGF-AA, sCD23 , SCF, TNF, LT-α, VEGF; rat IL-2, IL4, IL-6, IL-10, GM-CSF, IFN-γ, TNF).

Note 2: This ELISA pair is recommended primarily for measuring cytokine from experimental cell culture systems. These ELISAreagents are not recommended for assaying serum samples. For testing human IL-10 in complex biological fluids like serum or plasma, our human IL-10 OptEIA™ ELISA Set (Cat. No. 555157) and OptEIA™ ELISA Kit (Cat. No. 550613) are recommended.

WB: The JES3-12G8 antibody has been reported to be useful for Western blotting.. Please note that this application is not routinely tested at BD Biosciences Pharmingen.

JES3-12G8

The JES3-12G8 antibody reacts with human interleukin-10 (IL-10) and viral IL-10. The immunogen used to generate the JES3-12G8 hybridoma was human IL-10. This is a neutralizing antibody.

This antibody is routinely tested by ELISA detection. Other applications were tested at BD Biosciences Pharmingen during antibody development only or reported in the literature.

Biotin
Biotin is a ubiquitous co-factor (also known as Vitamin B7) that has many properties that make it extremely useful for molecular biology. Biotin has an extremely high affinity for the Avidin family of proteins (Kd = 10-15 M), making it the perfect tool to link two molecules. Biotin labeled antibodies can be combined with any number of Avidin-conjugated probes in order to customize an assay to a particular need. This is especially useful in the case of magnetic cell separation using streptavidin/magnetic bead conjugates, or in the case of flow cytometry using streptavidin/fluorophore conjugates.
