Recommended Assay Procedures
ELISA Capture: The purified A5-7 antibody can be used as the capture antibody in a sandwich ELISA for measuring rat IL-10 protein levels in conjunction with the biotinylated A5-6 antibody (Cat. # 555084) as the detecting antibody and recombinant rat IL-10 (Cat. #555113) as the standard. This purified capture antibody should be pretitrated in the range of 1 to 4 µg/ml to determine its optimal concentration for ELISA. To obtain linear standard curves, doubling dilutions of rat IL-10 ranging from 2,000 to 5 pg/ml should be used. For specific methodology, please visit the protocols section or chapter on ELISA in the Immune Function Handbook, both of which are posted on our web site, www.bdbiosciences.com.
This ELISA pair is recommended primarily for measuring cytokine from experimental cell culture systems. These ELISA reagents are not recommended for assay of serum or plasma samples. For measuring Rat IL-10 in serum or plasma, our BD OptEIA™ Rat IL-10 ELISA Set (Cat. No. 555134) is specially formulated and recommended.
Product Notices
- Since applications vary, each investigator should titrate the reagent to obtain optimal results.
- Please refer to www.bdbiosciences.com/us/s/resources for technical protocols.
- Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.
The A5-7 antibody reacts with rat Interleukin-10 (IL-10). The immunogen used to generate this hybridoma was E. coli-expressed rat IL-10.
