Two-color flow cytometric analysis of CD205 expression on mouse bone marrow cells. C57BL/6 mouse bone marrow cells were harvested and erythrocytes were lysed with BD Pharm Lyse™ Lysing Buffer (Cat. No. 555899). After washing, the bone marrow cells were preincubated with Purified Rat Anti-Mouse CD16/CD32 antibody (Mouse BD Fc Block™) (Cat. No. 553141/553142), and they were stained with APC Rat Anti-Mouse Ly-6G and Ly-6C antibody (Cat. No. 553129/561083) and either BD Horizon™ BV421 Rat IgG2a, κ Isotype Control (Cat. No. 562602; Left Plot) or BD Horizon BV421 Rat Anti-Mouse CD205 (DEC-205) antibody (Cat. No. 566375/566376; Right Plot) at 0.125 μg/test. Two-color flow cytometric contour plots showing the correlated expression of Ly-6G and Ly-6C versus CD205 (or Ig Isotype control staining) were derived from gated events with the forward and side light-scatter characteristics of viable cells. Flow cytometric analysis was performed using a BD LSRFortessa™ X-20 Cell Analyzer System. Data shown on this Technical Data Sheet are not lot specific.

Recommended Assay Procedures
For optimal and reproducible results, BD Horizon Brilliant™ Stain Buffer should be used anytime BD Horizon Brilliant™ dyes are used in a multicolor flow cytometry panel. Fluorescent dye interactions may cause staining artifacts which may affect data interpretation. The BD Horizon Brilliant Stain Buffer was designed to minimize these interactions. When BD Horizon Brilliant Stain Buffer is used in in the multicolor panel, it should also be used in the corresponding compensation controls for all dyes to achieve the most accurate compensation. For the most accurate compensation, compensation controls created with either cells or beads should be exposed to BD Horizon Brilliant Stain Buffer for the same length of time as the corresponding multicolor panel. The V18-949 monoclonal antibody specifically recognizes CD205 which is also known as Dendritic and thymic Epithelial Cells, 205 kDa (DEC-205). CD205 (DEC-205) is a type 1 transmembrane glycoprotein that is encoded by Ly75 (lymphocyte antigen 75). CD205 (DEC-205) is highly expressed by both CD8+ and CD103+ dendritic cell subsets and Langerhans cells. It is variably expressed by some other dendritic cell types, Gr1+ (Ly-6G and Ly-6C) bone marrow cells, B cells, macrophages, granulocytes, and mucosal and thymic cortical epithelial cells. CD205 (DEC-205) belongs to the C-type lectin receptor family and functions as a pattern recognition receptor and an endocytic receptor that participates in the cellular processing and presentation of foreign or self antigens to initiate immunity or tolerance.
