Pre-Optimized Detectors, No Voltage Adjustments
The system is equipped with a blue and a red laser, two light scatter detectors, and four fluorescence detectors with optical filters optimized for the detection of fluorochromes such as FITC, PE, PerCP, and APC. A compact optical design, fixed alignment, and pre-optimized detector settings make the system easier to use.
Data is digitally collected over a 7-decade dynamic range (16 million channels of digital data), making all data available to users as needed. Gating strategies and fluorescence compensation values can be set before, during, or after data collection. After data is collected, the BD Accuri CFlow® software Zoom function allows visualization of data at any scale, so that users can precisely set gates and regions.
Should updates in the values be required later, or if optimization is needed, simply change the settings and re-analyze the data. This flexibility also allows data to be re-examined to accommodate new research findings.
The system has been put through intense testing to ensure the design can withstand rugged conditions. Provided the system is anchored, it can run samples even if the benchtop is in motion, for example, onboard a ship.
T-cell phenotyping, 4-color analysis
Thawed human PMBCs were stained with directly labeled anti-CD45RA FITC, CD4 PE, CD8 PE-Cy7, and CD3 APC in PBS + 1 mg/mL of BSA, covered, on ice, for 30 minutes. Analysis was performed on the BD Accuri C6.
Selectable Lasers Module
The detectors and lasers of the standard BD Accuri C6 flow cytometer operate in a pre-defined configuration such that detectors FL1, FL2, and FL3 read blue laser-excited fluorescence emissions while detector FL4 reads red laser-excited emissions. This configuration is referred to as “3-blue 1-red”.
Installation of the optional Selectable Lasers Module on the BD Accuri C6 allows operation of the system in 2-blue/2-red and 4-blue configurations which significantly expands the fluorochrome combinations that can be analyzed. The 2-blue/2-red configuration can analyze FL1- and FL2-read blue laser-excited emissions and FL3- and FL4-read red laser-excited emissions. The 4-blue configuration detects blue laser-excited emissions.
The Selectable Lasers Upgrade requires installation of BD CFlow Plus or the BD CSampler which includes the selectable lasers activation key and 780/60, 610/20, 630/30 optical filters.
