Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with BD Horizon BB700 under optimal conditions that minimize unconjugated dye and antibody.

Recommended Assay Procedures

For optimal and reproducible results, BD Horizon Brilliant Stain Buffer should be used anytime two or more BD Horizon Brilliant dyes (including BD OptiBuild Brilliant reagents) are used in the same experiment.  Fluorescent dye interactions may cause staining artifacts which may affect data interpretation.  The BD Horizon Brilliant Stain Buffer was designed to minimize these interactions.  More information can be found in the Technical Data Sheet of the BD Horizon Brilliant Stain Buffer (Cat. Product Notices

  1. This antibody was developed for use in flow cytometry.
  2. The production process underwent stringent testing and validation to assure that it generates a high-quality conjugate with consistent performance and specific binding activity. However, verification testing has not been performed on all conjugate lots.
  3. Researchers should determine the optimal concentration of this reagent for their individual applications.
  4. An isotype control should be used at the same concentration as the antibody of interest.
  5. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

The 125518 monoclonal antibody specifically recognizes Mer. Mer is a single-pass type I transmembrane glycoprotein that is encoded by MERTK (MER proto-oncogene, tyrosine kinase) and is also known as MER, RP38, or STK kinase. Mer is comprised of an extracellular region with two immunoglobulin (Ig)-like domains and two fibronectin type III (FNIII) domains, a transmembrane segment and a conserved intracellular tyrosine kinase domain. Tyro-3, Axl, and Mer constitute the TAM subfamily of receptor tyrosine kinases (RTK). Mer is variably expressed by multiple cell types including monocytes, macrophages, dendritic cells, NK cells, platelets, and epithelial cells including retinal pigment epithelial cells. Mer binds to the vitamin K-dependent Growth arrest-specific protein 6 (Gas6) and to Protein S (ProS) through its extracellular Ig-like domains. Ligand binding leads to receptor dimerization, and autophosphorylation of tyrosine residues within the cytoplasmic Mer domains. This results in the activation of downstream signaling pathways that control cellular adhesion, aggregation, phagocytosis/efferocytosis, proliferation, survival, and migration. Through these activities, Mer plays major roles involved in development and in the regulation of hematopoiesis and immunity. Abnormal expression of Mer has been observed in various cancers and tumor cell lines.

The BD Horizon Brilliant™ Blue 700 (BB700) Dye is part of the BD Horizon Brilliant™ Blue family of dyes. This tandem fluorochrome is comprised of a polymer-technology dye donor with an excitation maximum (Ex Max) of 476-nm and an acceptor dye with an emission maximum (Em Max) at 695-nm. Driven by BD innovation, BB700 is designed to be excited by the blue laser (488-nm) and detected using an optical filter centered near 695-nm (e.g., a 695/20-nm bandpass filter). The donor dye can be excited by the Violet (405 nm) laser and the acceptor dye can be excited by the red (627–640 nm) laser resulting in cross-laser excitation and fluorescence spillover. BB700 Reagents are significantly brighter than equivalent PerCP or PerCP-Cy5.5 reagents and are less sensitive to photobleaching. In addition, BB700 shows much less excitation by the violet (407-nm) laser resulting in less spillover. BB700 has minimal yellow green (562-nm) excitation and is ideal for instruments with both blue (488-nm) and yellow green (562-nm) lasers. Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
