Preparation And Storage

The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. Store undiluted at 4°C.

Recommended Assay Procedures

ELISA Capture: The purified monoclonal antibody 20C2 (Cat. No. 555065) is useful as a capture antibody in a sandwich ELISA for measuring human IL-12 protein levels. Purified monoclonal antibody 20C2 can be paired with the biotinylated monoclonal antibody C8.6 (Cat. No. 554660) with recombinant human IL-12 p70 (Cat. No. 554613) as the standard. Purified monoclonal antibody 20C2 should be titrated to determine its optimal concentration for ELISA capture (2.0 µg/ml). To obtain linear standard curves, doubling dilutions of human IL-12 p70 ranging from ~2000 to 15 pg/ml are recommended for inclusion in each ELISA plate. Note 1: This ELISA pair is recommended primarily for measuring cytokines produced by experimental cell culture systems. These ELISA reagents are not recommended for measuring serum samples. The BD OptEIA™ Human IL-12 (p70) ELISA set (Cat. No. 555183) and Human IL-12 (p70) ELISA Kit (Cat. No. 559258) are specially-formulated for serum or plasma cytokine measurement.  

Note 2: This ELISA pair shows no cross-reactivity with any of the cytokines or chemokines tested  (including human IL-1α, IL-1β, IL-2, IL-3, IL-4, IL-5, IL-6, IL-7, IL-8, IL-9, IL-10, IL-11, IL-13, IL-15, IL-16, eotaxin, G-CSF, GM-CSF, GROα, GROβ, GROγ, IFN-γ, IP-10, lymphotactin, MCP-1, MCP-2, MCP-3, MCP-4, MIP-1α, MIP-1β, NAP-2, PF-4, RANTES, TNF, LT-α).

The 20C2 antibody reacts with the biologically active form of human IL-12 p70 heterodimer, but not with the p40 monomer or homodimer.

This antibody is routinely tested by ELISA Capture. Other applications were tested at BD Biosciences Pharmingen during antibody development only or reported in the literature.

Tissue culture supernatant is purified by either protein A/G or affinity purification methods. Both methods yield antibody in solution that is free of most other soluble proteins, lipids, etc. This format provides pure antibody that is suitable for a number of downstream applications including: secondary labeling for flow cytometry or microscopy, ELISA, Western blot, etc.
