Analysis of Zbtb46 Expression

     Left Panel - Two-color flow cytometric analysis Zbtb46 expression in mouse splenocytes. Mouse splenocytes were fixed and permeabilized using the BD Pharmingen™ Transcription Factor Buffer Set (Cat. No. 562574/562725). The cells were stained with BD Horizon™ BV421 Armenian Hamster Anti-Mouse CD11c (Cat. No. 565452) and either PE Rat IgG1, κ Isotype Control (Cat. No. 554685; Left Plot) or PE Rat Anti-Mouse Zbtb46 antibody (Cat. No. 565832; Right Plot). Two-color flow cytometric contour plots showing the correlated expression of Zbtb46 (or Ig Isotype control staining) versus CD11c were derived from gated events with the forward and side light-scatter characteristics of intact splenocytes.

No. 562367), and either PE Rat IgG1, κ Isotype Control (dashed line histogram) or PE Rat Anti-Mouse Zbtb46 antibody (solid line histogram).  The fluorescence histogram showing Zbtb46 expression (or Ig Isotype control staining) for conventional dendritic cells (cDC) was derived from CD11c-hi I-A/I-E-hi gated events with the forward and side light-scatter characteristics of intact splenic leucocytes.

    Flow cytometric analysis was performed using a BD FACSCanto™ II Flow Cytometer System.

Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with R-PE under optimum conditions, and unconjugated antibody and free PE were removed.

The U4-1374 monoclonal antibody specifically recognizes the Zinc finger and BTB domain containing 46 (Zbtb46) transcription factor, which is also known as BTB-ZF protein expressed in effector lymphocytes (BZEL), or BTB (POZ) domain containing 4 (Btbd4). Zbtb46 is a transcription factor that is selectively expressed by conventional dendritic cells (cDC; also known as classical dendritic cells) and their committed progenitors. It is not expressed by plasmacytoid DCs (pDC), monocytes, macrophages, or other lymphoid or myeloid lineages. Although Zbtb46 is not required for cDC development, it represses the expression of alternative myeloid growth factor receptors and thereby enforces cDC lineage restriction.

R-Phycoerythrin (PE), is part of the BD family of Phycobiliprotein dyes. This fluorochrome is a multimeric fluorescent phycobiliprotein with excitation maximum (Ex Max) of 496 nm and 566 nm and an emission maximum (Em Max) at 576 nm. PE is designed to be excited by the Blue (488 nm), Green (532 nm) and Yellow-Green (561 nm) lasers and detected using an optical filter centered near 575 nm (e.g., a 575/26-nm bandpass filter). As PE is excited by multiple lasers, this can result in cross-laser excitation and fluorescence spillover on instruments with various combinations of Blue, Green, and Yellow-Green lasers. Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
