Preparation And Storage

The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with FITC under optimum conditions, and unreacted FITC was removed. Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze.

Recommended Assay Procedures

Other applications: The purified format of the F11 clone has been reported to be useful in immunoprecipitation and IHC on frozen or zinc-fixed sections. Please note that these applications are not tested at BD Biosciences Pharmingen. This clone is also available in the no azide/low endotoxin format (NA/LE)  as Cat. No. 554950.

F11

The F11 monoclonal antibody specifically recognizes integrin β3 chain (CD61), which associates with the integrin αv chain (CD51), to form the vitronectin receptor found on endothelial cells, myeloid cells, and osteoclasts, and with the αIIb chain (CD41) on platelets and megakaryocytes.  Both receptors mediate adhesion to fibronectin, fibrinogen, vitronectin, thrombospondin, and von Willebrand factor.  The F11 antibody reportedly blocks the in vitro attachment of osteoclasts to several ligands.  Weak F11 antibody reactivity with human osteoclasts, megakaryocytes, and platelets has also been reported.

Fluorescein (FITC) is part of the BD blue family of dyes. This is a small organic fluorochrome with an excitation maximum (Ex Max) at 494-nm and an emission maximum (Em Max) at 518-nm. FITC is designed to be excited by the Blue laser (488-nm) and detected using an optical filter centered near 520 nm (e.g., a 530/30-nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
