Multi-color flow cytometric analysis of DCAR2 (Clec4b1) expression on mouse leucocytes and transfectants. Left and Middle Panels: Bone marrow cells from C57BL/6 mice were incubated in the presence of Purified Rat Anti-Mouse CD16/CD32 (Mouse BD Fc Block™) (Cat. No. 553141 or 553142) and were stained simultaneously with APC Hamster Anti-Mouse CD11c (Cat. No. 550261 or 561119) and FITC Rat Anti-Mouse I-A/I-E antibody (Cat. No. 553623 or 562009) to gate on conventional dendritic cells (cDCs) as shown in the left panel. At the same time, the samples of the bone marrow cells were also stained with either PE Rat Anti-Mouse DCAR2 (Clec4b1) (Cat. No. 566882) or PE Rat IgG2b, κ Isotype Control (Cat. No. 553989) at 0.5 µg/test. The overlayed plots of the middle panel were derived from gated cDCs showing expression of DCAR2 (Clec4b1) or Ig Isotype Control staining (solid and dashed line histograms, respectively). Right Panel: Non-transfected (dotted line histogram) and mouse DCAR2-transfected (solid line histogram) 293F cells were surface stained with PE Rat Anti-Mouse DCAR2 (Clec4b1) antibody (Cat. No. 566882) at 0.5 µg/test. Histogram plots showing expression of DCAR2 (Clec4b1) on non-transfected or Mouse DCAR2-transfected 293F cells were derived from gated events with forward and side light-scatter characteristics of viable cells. Flow cytometric analysis was performed using a BD LSRFortessa™ X-20 Flow Cytometer System and FlowJo™ software. Data shown on this Technical Data Sheet are not lot specific.

The 7D2D3 monoclonal antibody specifically recognizes mouse Dendritic Cell Activating Receptor 2 (DCAR2), a C-type lectin encoded by the Clec4b1 gene. DCAR2 (Clec4b1) is a member of the dendritic cell immunoreceptor (DCIR) family, which contains four dendritic cell inhibitory receptors (DCIR1-4) and two activating receptors (DCAR1, 2). The extracellular region of DCAR2 (Clec4b1) has one C-type lectin carbohydrate recognition domain (CRD), that is highly similar to that of DCIR1, followed by a transmembrane region. Instead of a cytoplasmic Immunoreceptor Tyrosine-based Inhibitory Motif (ITIM) that can transmit inhibitory signals, DCAR2 associates with the Immunoreceptor Tyrosine-based Activating Motif (ITAM)-containing adaptor molecule, Fc Receptor γ (FcRγ). DCAR2 (Clec4b1) is expressed on restricted subsets of conventional dendritic cells in bone marrow and skin-draining lymph nodes. C-type lectins bind to carbohydrates in a calcium-dependent manner, and this receptor may be involved in the regulation of immune responses to bacterial infections.
