Multi-parameter flow cytometric analysis of CD181 (CXCR1) expression on mouse blood neutrophils (left and middle panel). Mouse blood cells were stained simultaneously with APC Rat Anti-Mouse Ly-6G antibody (Cat. No. 560599), BD Horizon™ BV421 Rat Anti-Mouse CD11b antibody (Cat. No. 562605), and either PE Rat IgG2b, κ Isotype Control (Cat. No. 555848) or PE Rat Anti-Mouse CD181 (CXCR1) antibody (Cat. No. 566383) at 0.5 µg/test. Erythrocytes were then lysed with BD Pharm Lyse™ Lysing Buffer (Cat. No. 555899). The left panel displays two-color flow cytometric plots showing the correlated expression patterns of CD11b versus Ly-6G from gated events with the forward and side light-scatter characteristics of viable cells. The middle panel displays the fluorescence histograms of CD181 (CXCR1) expression (solid line histogram) or Ig Isotype control staining (dashed line histogram) on gated Ly-6G and CD11b double-positive neutrophils. Blood leukocytes that were Ly-6G and CD11b double-negative did not express CD181 (data not shown). Flow cytometric analysis of CD181 (CXCR1) expression on transfected 293F cells (right panel). Untransfected (dashed line histogram) and mouse CD181-transfected (solid line histogram) 293F cells were stained with PE Rat Anti-Mouse CD181 (CXCR1) antibody (Cat.No. 566383) at 0.5 ug/test. The fluorescence histogram showing CD181 (CXCR1) expression was derived from gated events with the forward and side light-scatter characteristics of viable cells. Flow cytometric analysis was performed on a BD LSRFortessa™ X-20 Flow Cytometer System. Data shown on this Technical Data Sheet are not lot specific.

The U45-632 monoclonal antibody specifically binds to mouse CD181, also known as C-X-C chemokine receptor type 1 (CXCR1) or IL-8 receptor α. CD181 is a chemokine receptor of the G-protein coupled receptor 1 family, also known as seven-transmembrane domain receptors. In the mouse, which is not known to express IL-8 (CXCL8), CD181 binds to mouse GCP-2 (CXCL6). Mouse CD181 can also bind to human IL-8. CD181 is expressed by neutrophils, which are activated upon ligand binding. The chemokine receptors CXCR1 and CXCR2 play an important role in mediating neutrophil recruitment and neutrophil-dependent injury in several models of inflammation.
