Flow cytometric analysis of CD204 (SR-AI, MSR1) expression on mouse monocyte/macrophage tumor cells and normal mouse peritoneal cavity cells (PEC). Left Plot: RAW 264.7 cells (ATCC TIB-71) were detached using trypsin and preincubated with Purified Rat Anti-Mouse CD16/CD32 antibody (Mouse BD Fc Block™) (Cat. No. 553141 or 553142). The cells were then stained with either BD Horizon™ BV421 Rat IgG2b, κ Isotype Control (Cat. No. 562603; dashed line histogram) or BD OptiBuild™ BV421 Rat Anti-Mouse CD204 (SR-AI, MSR1) (Cat. No. 748083; solid line histogram) at 0.25 µg/test. The fluorescence histograms showing CD204 (SR-AI, MSR1) [or Ig Isotype control] staining were derived from gated events with the forward and side light-scatter characteristics of viable singlet cells. Middle and Right Plots: PEC from BALB/c mice were preincubated with Purified Rat Anti-Mouse CD16/CD32 antibody (Mouse BD Fc Block™) (Cat. No. 553141 or 553142). The cells were then stained with Alexa Fluor® 647 Rat Anti-Mouse F4/80 (Cat. No. 565853 or 565854) and either BD Horizon™ BV421 Rat IgG2b, κ Isotype Control (Cat. No. 562603; Middle Plot) or BD OptiBuild™ BV421 Rat Anti-Mouse CD204 (SR--AI, MSR1) (Cat. No. 748083; Right Plot) at 0.25 μg/test. The pseudocolor dot plots showing CD204 (SR-AI, MSR1) [or Ig Isotype Contro]) staining versus F4/80 were derived from gated events with the forward and side light-scatter characteristics of intact singlet leucocytes. Flow cytometry and data analysis were performed using a BD LSRFortessa™ X-20 Cell Analyzer System and FlowJo® software. Recommended Assay Procedures
For optimal and reproducible results, BD Horizon Brilliant Stain Buffer should be used anytime two or more BD Horizon Brilliant dyes (including BD OptiBuild Brilliant reagents) are used in the same experiment. Fluorescent dye interactions may cause staining artifacts which may affect data interpretation. The BD Horizon Brilliant Stain Buffer was designed to minimize these interactions. The 268318 monoclonal antibody specifically recognizes CD204 which is also known as Scavenger receptor class A member 1 (SR-A1 or SCARA1), Scavenger receptor type A (SRA-A), Macrophage acetylated LDL receptor I and II, or Macrophage scavenger receptor types I and II. CD204 is a trimeric type II transmembrane glycoprotein that is encoded by MSR1 (Msr1 macrophage scavenger receptor 1) which belongs to the type 1 class A scavenger receptor family. The C-terminal extracellular region of CD204 contains a scavenger receptor cysteine-rich domain followed by a collagen domain and an alpha-helical coiled coil. It is expressed on monocytes, macrophages, microglia, dendritic cells, and mast cells. CD204 is involved in the uptake of various negatively-charged macromolecules including modified proteins, polysaccharides, polyribonucleic acids, and microbial products and plays a role in innate and acquired immunity.
The antibody was conjugated to BD Horizon™ BV421 which is part of the BD Horizon Brilliant™ Violet family of dyes. With an Ex Max of 407-nm and Em Max at 421-nm, BD Horizon BV421 can be excited by the violet laser and detected in the standard Pacific Blue™ filter set (eg, 450/50-nm filter). BD Horizon BV421 conjugates are very bright, often exhibiting a 10 fold improvement in brightness compared to Pacific Blue conjugates.
