Multicolor flow cytometric analysis of CD96 (TACTILE) expression on mouse splenocytes. C57BL/6 mouse splenic leucocytes were preincubated with Purified Rat Anti-Mouse CD16/CD32 antibody (Mouse BD Fc Block™) (Cat. No. 553141/553142). The cells were washed and then stained with Alexa Fluor® 647 Rat Anti-Mouse CD3 antibody (Cat. No. 557869; Top Plots), PerCP-Cy™5.5 Mouse Anti-Mouse NK1.1 antibody (Cat. No. 551114/561111; Bottom Plots) and either BD Horizon™ BV421 Rat IgG2a, λ Isotype Control (Cat. No. 562965; Left Plots) or BD Horizon BV421 Rat Anti-Mouse CD96 (TACTILE) antibody (Cat. No. 565687; Right Plots). Two-color flow cytometric contour plots showing the correlated expression of CD3 or NK 1.1 versus CD96 (TACTILE) [or Ig Isotype Control staining] were derived from gated events with the forward and side light-scatter characteristics of viable splenic leucocytes. Flow cytometric analysis was performed using a BD LSRFortessa™ X-20 Cell Analyzer System.

Recommended Assay Procedures
For optimal and reproducible results, BD Horizon Brilliant Stain Buffer should be used anytime two or more BD Horizon Brilliant dyes are used in the same experiment. Fluorescent dye interactions may cause staining artifacts which may affect data interpretation. The BD Horizon Brilliant Stain Buffer was designed to minimize these interactions. The 6A6 monoclonal antibody specifically binds to mouse CD96, which is also known as T cell-activated increased late expression protein (Tactile, TACT). CD96 is a type I transmembrane glycoprotein that belongs to the Ig gene superfamily. It is variably expressed on NK cells, and T cells at all developmental stages, including thymocytes and mature αβ T cells, γδ T cells, and NKT cells. CD96 interacts with Nectin-1 (CD111) and CD155 (poliovirus receptor). CD96 can reportedly mediate adhesive interactions between NK cells or T cells and CD155+ target cells. CD96 may compete with CD226 (DNAM 1) for CD155 binding and thus limit NK cell cytokine responses.
The antibody was conjugated to BD Horizon BV421 which is part of the BD Horizon Brilliant™ Violet family of dyes. With an Ex Max of 407-nm and Em Max at 421-nm, BD Horizon BV421 can be excited by the violet laser and detected in the standard Pacific Blue™ filter set (eg, 450/50-nm filter). BD Horizon BV421 conjugates are very bright, often exhibiting a 10 fold improvement in brightness compared to Pacific Blue conjugates.
