Description
Anti–HLA-DR, clone L243, is derived from the hybridization of mouse NS-1/1-Ag4 myeloma cells with spleen cells from BALB/c mice immunized with the human lymphoblastoid B-cell line RPMI 8866. CD14, clone MφP9, is derived from hybridization of mouse Sp2/0 myeloma cells with spleen cells from BALB/c mice immunized with peripheral blood monocytes from a patient with rheumatoid arthritis. Anti–HLA-DR recognizes a human class II major histocompatibility complex (MHC) antigen. The antigen is a transmembrane glycoprotein composed of α and β subunits that have molecular weights of 36 and 27 kDaltons (kDa), respectively. Anti–HLA-DR reacts with a nonpolymorphic HLA-DR epitope and does not cross-react with HLA-DQ or HLA-DP molecules. Anti-Monocyte PerCP-Cy5.5 reagent recognizes monocytes using two discrete specificities. This reagent includes CD14, a human monocyte/macrophage antigen, Mr 55 kDa, that recognizes the majority of the monocyte population. In addition, Anti-Monocyte PerCP-Cy5.5 recognizes FcR1 (CD64) by taking advantage of the unique recognition of CD64 by cyanine dyes. Anti-Monocyte PerCP-Cy5.5 is specially formulated to take advantage of this property of the cyanine dye to ensure detection of all monocytes (CD14 brightly positive and weakly positive) in a sample.
Preparation And Storage
The Anti–HLA-DR/Anti-Monocyte PerCP-Cy5.5 reagent is supplied as a combination of Anti–HLA-DR PE (≥95% 1:1 PE:mAb ratio) and Anti-Monocyte PerCP-Cy5.5 in a buffered gelatin solution containing an HLA-DR stabilizing reagent and 0.1% sodium azide. Store vials in the dark at 2° to 8°C. Do not freeze QuantiBRITE reagents; protect them from prolonged exposure to light. Each reagent is stable for the period shown on the bottle label when stored as directed.
