Description
The reagent is provided in 1 mL of phosphate-buffered saline (PBS) containing bovine serum albumin (BSA) and 0.1% sodium azide. The reagent contains FITC-labeled CD45, clone 2D1, and PE-labeled CD34, clone 8G12. The CD45/CD34 antigen recognizes a 180 to 220-kilodalton (kDa) human leucocyte antigen that is a member of the leucocyte common antigen (LCA) family. The CD45/CD34 antigen is present on all human leucocytes and is weakly expressed on hematopoietic progenitor cells. CD34 recognizes the human progenitor cell antigen (HPCA). The CD34 antigen is present on immature hematopoietic precursor cells and all hematopoietic colony-forming cells in bone marrow and blood, including unipotent and pluripotent progenitor cells. The CD45 and CD34 antibodies are each composed of mouse γ1 heavy chains and kappa light chains. BD TruCOUNT™ Tubes can be used with this reagent. Add the reagent and the blood sample directly to the BD TruCOUNT Tube to directly determine the absolute count of the cell population of interest. Simply divide the number of cellular events by the number of bead events, and then multiply by the bead concentration.
Preparation And Storage
Store the reagent vials upright at 2° to 8°C to maintain stability. Keep the vials dry and protected from light. Do not use after the expiration date shown on the label.
Store BD TruCOUNT Tubes in the foil pouch at room temperature (20° to 25°C) and use within 1 hour after removal from the foil pouch. Reseal the foil pouch immediately after each use. The tubes are stable in the closed pouch for the period shown on the pouch label. Once the pouch has been opened, do not use the tubes beyond the expiration date indicated on the packaging.
• Do not freeze the reagents or expose them to direct light during storage or during incubation with cells.
• Do not use the reagents if you observe any changes in appearance. Precipitation or a change in color indicates instability or deterioration.
• Examine the desiccant each time you open the foil pouch to use a BD TruCOUNT Tube. If the desiccant has turned from blue to lavender, discard the remaining tubes.
