Flow cytometric analysis of CD151 expression on mouse brain endothelioma cells. bEnd.3 cells (ATCC CRL-2299) were harvested using Cell Dissociation Buffer (Gibco 13151014), and then stained with either BD Horizon™ BV421 Rat IgG2a, κ Isotype Control (Cat. No. 562602; dashed line histogram) or BD OptiBuild™ BV421 Rat Anti-Mouse CD151 (Cat. No. 748109; solid line histogram) at 1.0 µg/test. The fluorescence histograms showing CD151 [or Ig Isotype control] staining were derived from gated events with the forward and side light-scatter characteristics of viable singlet cells. Flow cytometry and data analysis were performed using a BD LSRFortessa™ X-20 Cell Analyzer System and FlowJo™ software. Recommended Assay Procedures
For optimal and reproducible results, BD Horizon Brilliant Stain Buffer should be used anytime two or more BD Horizon Brilliant dyes (including BD OptiBuild Brilliant reagents) are used in the same experiment. Fluorescent dye interactions may cause staining artifacts which may affect data interpretation. The BD Horizon Brilliant Stain Buffer was designed to minimize these interactions. The 455807 monoclonal antibody specifically recognizes CD151, which is also known as Platelet-Endothelial Tetraspan Antigen 3 (PETA-3). CD151 is a member of the Tetraspanin or Transmembrane 4 superfamily (TM4SF). Like most tetraspaninins, CD151 regulates cell migration, interactions, and signaling by forming complexes with other membrane molecules, such as integrins and other tetraspanins. CD151 is expressed on many types of cells, including endothelial and epithelial cells, megakaryocytes and platelets, but not on lymphoid cells.
The antibody was conjugated to BD Horizon™ BV421 which is part of the BD Horizon Brilliant™ Violet family of dyes. With an Ex Max of 407-nm and Em Max at 421-nm, BD Horizon BV421 can be excited by the violet laser and detected in the standard Pacific Blue™ filter set (eg, 450/50-nm filter). BD Horizon BV421 conjugates are very bright, often exhibiting a 10 fold improvement in brightness compared to Pacific Blue conjugates.
