Preparation And Storage

The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. Store undiluted at 4°C.

Recommended Assay Procedures

G146-222 has been shown to recognize in vitro translated, recombinant bacterially expressed and endogenous B cell Fli-1. In gel shift assays, this antibody disrupts complexes of Fli-1 and DNA probe, however, this application is not tested at BD Pharmingen.

Fli-1 is a 50 kDa ets-related transcription factor. Like other ets-related proteins it binds to consensus sites in DNA through an 85 amino acid ets domain in the carboxyl region of the protein.  Fli-1 is associated with Ewing sarcoma (EWS) through a t(11;22)(q24;q12) chromosomal translocation. This translocation brings the 5' region of the EWS gene into conjunction with the 3' region of the Fli-1 gene encoding the ets-DNA binding domain. Such a translocation is found in 85% of Ewing sarcomas. The resulting fusion protein has the DNA binding activity of Fli- 1 and, with the EWS domain, it is also a more potent transcriptional activator than Fli-1 itself.  The strong transforming activity of the EWS-Fli-1 fusion protein may in part be due to its ability to transactivate the promoter for c-myc. G146-222 recognizes human Fli-1. The antibody was raised against a bacterially expressed Fli-1 ets domain fusion protein.
