Flow cytometric analysis of OX2R (CD200R1) expression on human peripheral blood leucocyte populations. Whole blood cells were stained with either BD Horizon™ BV421 Mouse IgG1, κ Isotype Control (Cat. No. 562438, Left Plot) or BD Horizon BV421 Mouse Anti-Human OX2R (CD200R1) antibody (Cat. No. 566344; Right Plot) at 1 μg/test. The erythrocytes were lysed with BD Pharm Lyse™ Lysing Buffer (Cat. No. 555899). The contour plots showing the expression of CD200R (or Ig Isotype control staining) were derived from gated events with the forward and side light-scatter characteristics of viable leucocytes. Flow cytometric analysis was performed using a BD™ Canto II Flow Cytometer System. Data shown on this Technical Data Sheet are not lot specific.

For optimal and reproducible results, BD Horizon Brilliant Stain Buffer should be used anytime two or more BD Horizon Brilliant dyes are used in the same experiment. Fluorescent dye interactions may cause staining artifacts which may affect data interpretation. The BD Horizon Brilliant Stain Buffer was designed to minimize these interactions. More information can be found in the Technical Data Sheet of the BD Horizon Brilliant Stain Buffer (Cat. No. 563794).
The OX-108 monoclonal antibody specifically binds to CD200R which is also known as OX2 receptor 1 (OX2R) or MOX2 receptor (MOX2R). CD200R is a type I transmembrane glycoprotein that is encoded by CD200R1 (CD200 receptor 1) and belongs to the C2-set domain containing gene family within the Ig superfamily. CD200R functions as an inhibitory receptor when bound by its ligand, CD200. CD200R is expressed by hematopoietic cells including monocytes, macrophages, dendritic cells, Langerhans cells, neutrophils, basophils, most T cells and mast cells. CD200R signaling may regulate the functions of various types of leucocytes.
The antibody was conjugated to BD Horizon BV421 which is part of the BD Horizon Brilliant™ Violet family of dyes. With an Ex Max of 407-nm and Em Max at 421-nm, BD Horizon BV421 can be excited by the violet laser and detected in the standard Pacific Blue™ filter set (eg, 450/50-nm filter). BD Horizon BV421 conjugates are very bright, often exhibiting a 10 fold improvement in brightness compared to Pacific Blue conjugates.
