Biotin Mouse Anti-Human CD57
Two-color flow cytometric analysis of CD57 expression on human peripheral blood lymphocytes. Whole blood was stained with APC Mouse Anti-Human CD3 antibody (Cat. No.561811) and with either Biotin Mouse IgM, κ Isotype Control (Cat. No. 553473; Left Plot) or Biotin Mouse Anti-Human CD57 antibody (Cat. No. 568064; Right Plot) at 1 µg/test. The cells were washed and counterstained with PE Strptavidin (Cat. No. 554061). Erythrocytes were lysed with BD FACS Lysing Solution (Cat. No. 349202). The bivariate pseudocolor density plot showing the correlated expression of CD57 (or Ig Isotype control staining) versus CD3 was derived from gated events with the forward and side light-scatter characteristics of intact peripheral blood lymphocytes. Flow cytometry and data analysis were performed using a BD LSRFortessa™ Cell Analyzer System and FlowJo™ software. Data shown on this Technical Data Sheet are not lot specific.

Preparation And Storage

Store undiluted at 4°C. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with biotin under optimum conditions, and unreacted biotin was removed.

NK-1

The NK-1 monoclonal antibody specifically reacts with a 110 kDa carbohydrate antigen associated with myelin-associated glycoprotein expressed on 7-35% of normal peripheral blood lymphocytes including a subset of natural killer cells, a subset of CD8-positive peripheral blood T cells, and on some neural tissues. CD57 is not expressed on granulocytes, platelets, red blood cells or thymocytes. The function of CD57 is still unclear, however, its expression on T-cell subets occurs in late immune responses.

Biotin
Biotin is a ubiquitous co-factor (also known as Vitamin B7) that has many properties that make it extremely useful for molecular biology. Biotin has an extremely high affinity for the Avidin family of proteins (Kd = 10-15 M), making it the perfect tool to link two molecules. Biotin labeled antibodies can be combined with any number of Avidin-conjugated probes in order to customize an assay to a particular need. This is especially useful in the case of magnetic cell separation using streptavidin/magnetic bead conjugates, or in the case of flow cytometry using streptavidin/fluorophore conjugates.
