Two-color flow cytometric analysis of human BTN3 (CD277) expression on human peripheral blood lymphocytes. Human whole blood was stained with APC Mouse Anti-Human CD3 antibody (Cat. No. 555335/561810/561811) and either PE Mouse IgG2a, κ Isotype Control (Cat. No. 553457; Left Plot) or PE Mouse Anti-Human CD277 antibody (Cat. No. 566006; Right Plot). Erythrocytes were lysed with BD Pharm Lyse™ Lysing Buffer (Cat. No. 555899). Two-color flow cytometric contour plots showing the correlated expression of BTN3 (CD277) [or Ig Isotype control staining] versus CD3 were derived for gated events with the forward and side light-scatter characteristics of viable lymphocytes. Flow cytometric analysis was performed using a BD FACSCanto™ II Flow Cytometer System.

- This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).

The 232-5 monoclonal antibody specifically recognizes Butyrophilin 3 (BTN3), which is also known as CD277. BTN3 is comprised of three structurally related members, BTN3A1, BTN3A2, and BTN3A3. BTN3 molecules are transmembrane type 1 glycoproteins that belong to the Ig superfamily. The 232-5 antibody recognizes the variable region of the extracellular domains of BTN3 molecules. BTN3 molecules are differentially expressed by T, NKT, B, and NK cells, monocytes, macrophages, dendritic cells, as well as some stem cells and tumor cells. BTN3 expression is decreased upon lymphocyte activation. The extracellular domain of CD277 is structurally related to the B7 family of proteins that regulate T cell activation. A soluble form of the 232-5 antibody can serve as an agonist in BTN3-mediated regulation of anti-CD3 ± anti-CD28 antibody activated CD4+ or CD8+ T cells.
