Two-color flow cytometric analysis of CD6 expression by mouse splenocytes. C57BL/6 mouse splenic leucocytes were stained with BD Horizon™ BV421 Hamster Anti-Mouse CD3e antibody (Cat. No. 562600) and either Alexa Fluor® 647 Rat IgG2a, λ Isotype Control (Cat. No. 564281; Left Plot) or Alexa Fluor® 647 Rat Anti-Mouse CD6 antibody (Cat. No. 566426; Right Plot) at 0.06 µg/test. A two-color flow cytometric contour plot showing the correlated expression of CD3e versus CD6 (or Ig Isotype control staining) was derived from gated events with the forward and side-light scattering characteristics of viable leucocytes. Flow cytometric analysis was performed using a BD FACSCanto™ II Flow Cytometer System. Data shown on this Technical Data Sheet are not lot specific.

Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated to Alexa Fluor® 647 under optimum conditions, and unreacted Alexa Fluor® 647 was removed.

Product Notices

  1. Since applications vary, each investigator should titrate the reagent to obtain optimal results.
  2. An isotype control should be used at the same concentration as the antibody of interest.
  3. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

The J90-462 monoclonal antibody specifically recognizes CD6. CD6 is a 105-130 kDa type I transmembrane glycoprotein that belongs to the scavenger receptor family. It is expressed on most thymocytes and peripheral T cells. CD6 is highly expressed on CD3-high thymocytes and expressed at lower levels on CD4+ CD8+ double positive and CD4+CD8- or CD4-CD8+ single positive thymocytes. CD6 is also reportedly expressed at low levels by some neurons.  CD6 serves as a receptor for CD166, also known as Activated Leukocyte Cell Adhesion Molecule (ALCAM). CD6 plays a role in adhesion and migration during organogenesis. The CD6 intracellular domain is constitutively phosphorylated which suggests that CD6 may play a signaling role in T cell activation.

Alexa Fluor™ 647 Dye is part of the BD red family of dyes. This is a small organic fluorochrome with an excitation maximum (Ex Max) at 653-nm and an emission maximum (Em Max) at 669-nm. Alexa Fluor 647 is designed to be excited by the Red laser (627-640 nm) and detected using an optical filter centered near 520-nm (e.g., a 660/20 nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
