Two-color flow cytometric analysis of human EMR2 (CD312) expression on human peripheral blood monocytes. Human whole blood was stained with PE Mouse Anti-Human CD14 antibody (Cat. No. 562691) and either Alexa Fluor® 647 Mouse IgG2b, κ Isotype Control (Cat. No. 557903; Left Panel) or Alexa Fluor® 647 Mouse Anti-Human EMR2 (CD312) antibody (Cat. No. 564342; Right Panel). Erythrocytes were lysed with BD Pharm Lyse™ Lysing Buffer (Cat. No. 555899). Two-color flow cytometric contour plots show the correlated expression patterns of EMR2 (CD312) (or Ig Isotype control staining) versus CD14 for gated events with the forward and side light-scatter characteristics of viable monocytes. Flow cytometric analysis was performed using a BD FACSCanto™ II Flow Cytometer System.

Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated to Alexa Fluor® 647 under optimum conditions, and unreacted Alexa Fluor® 647 was removed.

  1. This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).

  8. Source of all serum proteins is from USDA inspected abattoirs located in the United States.

The 494025 monoclonal antibody specifically binds to EMR2 (Epidermal Growth Factor Module-containing Mucin-like Receptor 2) glycoprotein which is also known as CD312. EMR2 belongs to the Epidermal Growth Factor-seven Span Transmembrane (EFG-TM7) family. EMR2 is expressed by activated lymphocytes and myeloid cells. CD312 can bind to the glycosaminoglycans, dermatan sulfate and chondroitin sulfate. Upon cellular activation, CD312 expression increases and localizes to the leading edge of neutrophils and regulates their adhesion, migration and production of superoxide anion. CD312 expression increases as monocytes differentiate into macrophages but decreases upon differentiation into dendritic cells. EMR2 may also mediate some interactions between activated B cells that express chondroitin sulfate and activated dendritic cells, macrophages and T cells.

Alexa Fluor™ 647 Dye is part of the BD red family of dyes. This is a small organic fluorochrome with an excitation maximum (Ex Max) at 653-nm and an emission maximum (Em Max) at 669-nm. Alexa Fluor 647 is designed to be excited by the Red laser (627-640 nm) and detected using an optical filter centered near 520-nm (e.g., a 660/20 nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
