FITC Rat Anti-Mouse Vβ 10[b] TCR
Two-color analysis of the expression of Vβ 10[b] TCR on peripheral lymphocytes. C57BL/6 lymph node cells were incubated simultaneously with FITC Rat Anti-Mouse Vβ 10[b] TCR (Cat. No.553284), PE Rat Anti-Mouse CD4 (Cat. No. 553048/553049), and PE Rat Anti-Mouse CD8a (Cat. No. 553032/553033) monoclonal antibodies. The fluorescence contour plots were derived from gated events based on the forward and side light-scattering characteristics of viable lymphocytes. Flow cytometry was performed on a FACScan™.

Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with FITC under optimum conditions, and unreacted FITC was removed.

Recommended Assay Procedures

For flow cytometry of cell suspensions from peripheral lymphoid tissues, it is recommended that multicolor staining be performed to distinguish T lymphocytes from non-T cells.

Product Notices

  1. Since applications vary, each investigator should titrate the reagent to obtain optimal results.
  2. An isotype control should be used at the same concentration as the antibody of interest.
  3. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.

The B21.5 monoclonal antibody specifically recognizes the Vβ 10 T-Cell Receptor (TCR) of mice having the b haplotype (e.g., A, AKR, BALB/c, CBA, C3H/He, C57BL, C58, DBA/1, DBA/2) of the Tcrb gene complex. It does not react with mice having the a (eg, C57BR, C57L, SJL, SWR) or c (eg, RIII) Tcrb haplotype.

Fluorescein (FITC) is part of the BD blue family of dyes. This is a small organic fluorochrome with an excitation maximum (Ex Max) at 494-nm and an emission maximum (Em Max) at 518-nm. FITC is designed to be excited by the Blue laser (488-nm) and detected using an optical filter centered near 520 nm (e.g., a 530/30-nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
