FITC Rat Anti-Mouse Vβ 2 TCR
Two-color analysis of the expression of Vβ 2 TCR on peripheral T lymphocytes. SJL lymph node cells were incubated simultaneously with FITC-conjugated B20.6, PE-conjugated RM4-5 (anti-CD4, Cat. No. 553048/553049), and PE-conjugated 53-6.7 (anti-CD8a, Cat. No. 553032/553033) monoclonal antibodies. Flow cytometry was performed on a FACScan™ (BDIS, San Jose, CA).

Preparation And Storage

The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated with FITC under optimum conditions, and unreacted FITC was removed. Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze.

Recommended Assay Procedures

For flow cytometry of cell suspensions from peripheral lymphoid tissues, it is recommended that multicolor staining be performed to distinguish T lymphocytes from non-T cells.

B20.6

The B20.6 monoclonal antibody specifically recognizes the Vβ 2 T-cell Receptor of all strains tested. Vβ 2 TCR-bearing T lymphocytes are clonally eliminated in mice expressing I-E and exposed neonatally to milk-borne BALB/cV virus, a highly tumorigenic strain of MMTV. Exposure of adult mice to BALB/cV virus results in expansion of Vβ 2 TCR-bearing T cells, which is partially dependent upon presentation by I-E.

This antibody is routinely tested by flow cytometric analysis. Other applications were tested at BD Biosciences Pharmingen during antibody development only or reported in the literature.

Fluorescein (FITC) is part of the BD blue family of dyes. This is a small organic fluorochrome with an excitation maximum (Ex Max) at 494-nm and an emission maximum (Em Max) at 518-nm. FITC is designed to be excited by the Blue laser (488-nm) and detected using an optical filter centered near 520 nm (e.g., a 530/30-nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
