Purified Rat Anti-Mouse CD3 Molecular Complex
CD3 expression in spleen and thymus. BALB/c splenocytes were simultaneously stained with purified mAb 17A2 (bottom left panel) and PE-conjugated anti-mouse CD19 mAb 1D3 (top left & bottom left panels), followed by FITC-conjugated anti-rat IgG2b mAb G15-337 (Cat. No. 553884) (top left & bottom left panels). BALB/c thymocytes were also stained with 17A2 mAb (bottom right panel), followed by FITC-conjugated anti-rat IgG2b (top right & bottom right panels). Flow cytometry was performed on a BD FACSCan™ flow cytometry system.

Preparation And Storage

The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. Store undiluted at 4°C.

Recommended Assay Procedures

Precautions:  Polyclonal goat anti-rat Ig (Cat. No. 554016) has been reported to have weak reactivity to to this rat IgG2b mAb. Therefore, the use of FITC-conjugated anti-rat IgG2b mAb G15-337 (Cat. No. 553884) is recommended as a second step for flow cytometry.

Note:  The hamster anti-mouse CD3e mAb 145-2C11 (Cat. No. 553057) has been reported to induce greater functional responses than 17A2 antibody in assays which depend on presentation of the monoclonal antibody by Fc receptor-positive macrophages.

17A2

The 17A2 monoclonal antibody specifically binds to the T-cell receptor-associated CD3 complex that is expressed on many thymocytes and mature T lymphocytes. Plate-bound 17A2 antibody has been reported to induce IL-2 production by cultured T cells in the absence of accessory cells. The binding of the 17A2 antibody to T cells can be blocked by the anti-CD3e mAb 145-2C11 (Cat. No. 557306/553058/550275). This suggests that the 17A2 antibody recognizes an epitope of the CD3 epsilon chain. In vivo treatment with 17A2 antibody has been reported to partially deplete T lymphocytes and temporarily down-modulates CD3 expression on T cells.

This antibody is routinely tested by flow cytometric analysis. Other applications were tested at BD Biosciences Pharmingen during antibody development only or reported in the literature.

Tissue culture supernatant is purified by either protein A/G or affinity purification methods. Both methods yield antibody in solution that is free of most other soluble proteins, lipids, etc. This format provides pure antibody that is suitable for a number of downstream applications including: secondary labeling for flow cytometry or microscopy, ELISA, Western blot, etc.
