Immunohistofluorescent analysis of CD4 expression by cells within human tonsil (left panel). A human tonsil cryosection (5 µm) was fixed with BD Cytofix™ Fixation Buffer (Cat. No. 554655), blocked with 5% goat serum and 1% BSA diluted in 1x PBS, and stained with Alexa Fluor® 488 Mouse Anti-Human CD4 antibody (Cat. No. 557695, pseudo-colored red), BD Horizon™ BV421 Mouse Anti-Human CD19 antibody (Cat. No. 562440/562441, pseudo-colored green), and Alexa Fluor® 555 Mouse Anti-Human Ki-67 antibody (Cat. No. 558617, pseudo-colored yellow). The section was then counterstained with DRAQ5 (Cat. No. 564902/564903, pseudo-colored blue). Images were captured on a standard epifluorescence microscope. Preparation And Storage

Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze. The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography. The antibody was conjugated to Alexa Fluor® 488 under optimum conditions, and unreacted Alexa Fluor® 488 was removed.

Product Notices

  1. This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-µl experimental sample (a test).
  2. An isotype control should be used at the same concentration as the antibody of interest.
  3. Source of all serum proteins is from USDA inspected abattoirs located in the United States.
  4. Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before discarding to avoid accumulation of potentially explosive deposits in plumbing.
  5. Alexa Fluor® 488 fluorochrome emission is collected at the same instrument settings as for fluorescein isothiocyanate (FITC).

RPA-T4

The RPA-T4 monoclonal antibody specifically binds to CD4, a 59 kDa single-chain transmembrane glycoprotein that is expressed on T-helper/inducer cell populations. CD4 is also expressed on thymocyte subsets and at lower levels on monocytes and macrophages. CD4 functions as a co-receptor in MHC class II-restricted antigen-induced T cell activation and as a receptor for human immunodeficiency viruses (HIV). This antibody binds to the D1 domain (CDR1 and CDR3 epitopes) of the CD4 antigen and reacts with approximately 80% of thymocytes and 45% of peripheral blood lymphocytes. RPA-T4 is capable of blocking HIV-1, gp120, and inhibits syncytium formation.

Alexa Fluor™ 488 Dye is part of the BD blue family of dyes. This is a small organic fluorochrome with an excitation maximum (Ex Max) at 494-nm and an emission maximum (Em Max) at 517-nm. Alexa Fluor™ 488 is designed to be excited by the Blue laser (488 nm) and detected using an optical filter centered near 520-nm (e.g., a 530/30-nm bandpass filter). Please ensure that your instrument’s configurations (lasers and optical filters) are appropriate for this dye.
